Astral Bar at Star City Casino might be worth a visit. While not really a wine bar as such, you can order any of wines from the restaurant menu. Has amazing views across Darling Harbour to the city and if the weather is nice you can sit outside.

Wine bars seem to be cropping up every week, however here’s a couple of favourites.
1. De Vine (cnr of Clarence & Market, near QVB)
Andreas (owner) is generally there and I often let him choose the wine (within budget) as he often has a few crackers not on the wine list. If you like Euro Rieslings you’ll be happy here. The bar food is good, they have a restaurant also but I’ve never eaten there. Most wines generally have less than 100% mark up.
2. Bambini Trust (Elizabeth St, across from Hyde Park)
Only been here a few times but have enjoyed myself every time. Broad range and the food is v good.
If you’re staying in the Rocks, Wine Odyssey (Argyle St) is worth a look

Food-wise Ash St Cellars is the go I'm told. They have a chef's hat. The wine list looks interesting as well.

North Sydney Cellars gave me one of my best food and wine experiences of last year, hell, any year.
Cannot recommend it highly enough.

Ash Street could be very busy on the weekend, but the wine list is interesting.
I also like Fix St James and Glass at the Hilton.
Wine Odyssey is a worthwhile experience.

One of my favourites is Mille Vini on Crown St in Surry Hills. Great wine list, great food, and very cool, initimate atmosphere

Not a wine bar per se but for something completely different, an Absinthe bar has just opened up in Surry hills. You need to like the green stuff as that's all they serve but if you want an unusual expereince, then it could be of interest.

Indeed. I drank a bit of Absinthe in Prague and it did lead to some unusual experiences.

OMG I am so going to go there. I love Absinthe (I'll take a bag of Bassetts with me as well)

Thanks everyone for all the suggestions; I ended up at Mille Vini and De Vine, mostly because they were close to where I was staying and it wouldn't stop raining in Sydney! Both fantastic places and I wish Adelaide did this type of thing more. Both had great selections of Italian wines by the glass, with the highlight probably being a 2004 Barolo last night at De Vine.
Atmosphere, service and food comparably excellent at both; happily recommend either.
I'll just need to return to Sydney soon to try out the rest. In the meantime, if anyone wants to set up (or suggest) something similar in Adelaide, I'll be there soon and plenty.

Would anything like that work in Adelaide? I always get the impression when I'm back there that the parochial favouritism of SA wines would make it difficult for out-of-State focussed wine bars to work.
